I have seen Error 6 also appear when a user had a copy of FileMaker running that had the same serial/license number as another currently active user. Once the other user quit FileMaker, the Error 6 disappeared and the user could access files.

I've also had this happen when (stupidly) opening a file from the body of an email without first saving it to my computer. It might also even be open from an archive.

This generally happens after copying files from a CD. Especially prone to Mac users. Do a "get info" of the file and reset or turn off the read-only selection.
